Nobody knows yet who she is referring too. She could be referring to Isshin as being their Uncle atau she may be refering to Isshin as their Uncle because that is how Ganju remembered him not realizing he was of a different relation atau not related at all.
She could also be referring to Ukitake as uncle as she is talking with family as her rescuing the three Fullbringers brought them into the family and the Shiba clan could have thought of him as an uncle despite the fact they aren't related which would explain why they were on good terms with him despite him being a Shinigami, atau one of Ukitake's female siblings happened to be their mother.
It could also be a reference to the elder that Ichigo and his Friends meet up with in the Rukongai as he may have been the first to recognize that Ichigo was possibly related to the Shiba clan and asked Kukaku not to voice his theory atau something like that.
Her use of uncle does hint at the possibility that Isshin and Ichigo are likely related to the Shiba clan in some shape atau form, but don't consider it to be a confirmation that it is true.
